Regulation of Proinflammatory Cytokine Gene Transcription in Activated Macrophages
Stephen T. Smale, Ph.D.
Investigator,
University of California, Los Angeles, School of Medicine Biography... Summary: Stephen Smale studies mechanisms of transcriptional regulation during the development and function of the immune system. An inflammatory response can be initiated by the interaction between a microbial pathogen and host macrophages. This interaction leads to the transcriptional induction of several proinflammatory cytokine genes, which then generate the inflammatory response. Toll-like receptors (TLRs) comprise a family of mammalian cell-surface proteins that stimulate proinflammatory cytokine gene transcription in response to lipopolysaccharide (LPS) and other bacterial products. TLRs induce transcription by stimulating signal transduction pathways that ultimately lead to the induction of specific transcription factors.
Although several transcription factors involved in proinflammatory cytokine gene regulation have been identified, a number of observations suggest that our knowledge of the regulatory mechanisms for these genes remains at a preliminary stage. For example, several important agents that enhance and suppress inflammation by modulating proinflammatory cytokine gene transcription have little or no impact on the known regulators of these genes. Thus novel approaches will be needed to dissect the mechanisms underlying these immunoregulatory events.

Stephen Smale
Stephen Smale was born in Flint, Michigan. He attended the University of Michigan, obtaining his B.S. in 1952 and his M.S. the following year. Smale continued to work for his doctorate at the University of Michigan, Ann Arbor, and he was awarded his Ph.D. in 1957 for the thesis Regular Curves on Riemannian Manifolds . In his thesis he generalized results proved by Whitney in 1937 for curves in the plane to curves on an n-dimensional manifold. He became an instructor at the University of Chicago in 1956. He did an NSF postdoctoral fellowship at the Institute for Advanced Study at Princeton from 1958 to 1960. He taught math at the University of California at Berkeley, then at Columbia University, then returned to a professorship at the University of California at Berkeley where he has spent the main part of his career. He retired from Berkeley in 1995 and took up a post as professor at the City University of Hong Kong. By the late 1960's, Smale had moved into applications. He modelled physical processes by dynamical systems, opening new lines of inquiry. The n-body problem and electric circuit theory were among the applications that Smale framed in the language of dynamical systems. For much of the 1970's, he focused on economics, injecting topology and dynamics into the study of general economic equilibria. Having established the nature of equilibria, Smale began to think algorithms for their computation. Smale's recent work has been on theoetical computer science. With co-workers Blum and Shub, he has developed a model of computation which includes both the Turing machine approach and the numerical methods of numerical analysis.

Stephen Smale
From Wikipedia, the free encyclopedia. Stephen Smale is an American mathematician and winner of the Fields Medal . In 1998 he constructed a list of 18 problems in mathematics to be solved in the 21st century. This list was compiled in the spirit of Hilbert 's famous list of problems produced in 1900. In fact, Smale's list includes some of the original Hilbert problems. Smale's problems include the Jacobian conjecture and the Riemann hypothesis , both of which are still unsolved.

PhD (Michigan)
B orn in Michigan, USA, Professor Smale received his PhD degree from the University of Michigan in 1957, and within four years became a full Professor at Columbia University. In 1964, he was named a Professor at the University of California, Berkeley and held the post for 30 years before joining City University as a Distinguished University Professor. Professor Smale has made significant contributions in the fields of dynamical systems, geometry, econometrics, operational research, topology and the mathematical theory of computer science. These contributions have resulted in a number of academic awards and achievements including his holding of the prestigious Alfred Solan Research Fellowship from 1960-62. In 1966, Professor Smale won a Fields Medal - a international medal awarded once every four years for outstanding discoveries in mathematics. This honour is comparable to a Nobel Prize and is traditionally awarded to mathematicans under 40 years of age. Important honours bestowed upon Professor Smale during his distinguished academic career include the 1965 Veblen Prize for Geometry, awarded every five years by the American Mathematical Society; in 1988, the Chauvenet Prize by the Mathematical Association of America; and in 1989, the Von Neumann Award by the Society for Industrial and Applied Mathematics. Professor Smale is a member of both the National Academy of Sciences and the American Academy of Arts and Sciences. He is recognized internationly in many fields of Mathematics, and has been invited as Visiting Professor to such esteemed institutions as College de France, Paris (Spring 1962), University of Paris, Orsay (Fall 1972-73), Yale University (Fall 1974) and Columbia University (Fall 1987).
