Dan-Hui Yang On the Molecular Mechanism of LHCII Proteolysis During Photoacclimation of Photosystem II Higher plants and green algae have a remarkable ability to respond to fluctuations in the light intensity. Plants adapted to low or high light regimes possess larger or smaller light-harvesting antennae respectively. Although this long-term photoacclimation of the PSII antennae is an important factor for the photoprotection of photosynthetic apparatus and it has been extensively described at the physiological level, the underlying molecular mechanisms have remained largely unknown. Furthermore, the proteolytic system involved has been characterized in vitro and found not to correspond to any previously characterized thylakoid proteases. Purification of the novel proteolytic system from acclimating spinach leaves reveal that it is represented by a 44 kDa protease, probably a chloroplast homologue of the Lon protease in E. coli., which may consist of an ATPase/SSD domain and a protease domain. The fragmentation pattern during LHCII degradation in vitro gives support on requirement of multiple proteolytic cleavages for the complete digestion of LHCII. Finally, the mechanism of redox controlled turnover of LHCII during photoacclimation was studied using Lemna perpusilla wild type and a cytochrome b6f-deficient mutant as a model system. The results strongly suggest that plastoquinone, as an important redox regulator in signal transduction, affects both synthesis and degradation of LHCII during photoacclimation of PSII.

Chung-Hui (Rebecca) Yang's Home Page I am a PhD student in Biological sciences at Stanford working on Drosophila eye development. I was born in Taiwan and I came to the US after college because all my best friends ended up in the US. Shown below is a picture from my research. Quarterly Report for Fall ‘01 Hui Yang yanghu@cis.ohio-state.edu In this report, you will read the following: a summary of the work finished in the quarter of Fall 2001; a general description of all the files generated so far; and finally, a to-do list for the projects: newLIPIDAG and LIMP. Summary of Work finished in Quarter Fall’01 My main work in the last quarter involved three projects, they are: newLIPIDAG : the web enabled database of phase diagrams System migration/upgrading: migrating all our web sites from Windows NT4.0 to Windows 2000. LIMP: Lipid Physic-Chemical Properties Database NewLIPIDAG I started working on newLIPIDAG in the middle of Summer quarter in 2001. In Summer’01, I finished the entire coding task for data conversion (from Excel to MS Access2000) and integration (with the other databases, such as newLIPIDAT, LIMP and oldLIPIDAG). Please refer to my summer quarter’s report for more details. In the beginning of last Fall’01, I completed the debugging and testing on all the conversion programs; this work ensures all the newLIPIDAG data were converted correctly from the Excel format and, also, integrated correctly with the data from the other databases. Following the conversion stage, the next stage would be making all newLIPIDAG data web accessible, which was the core task in Fall’01.

The group members are highly recommended to give their feedback to HuiYang. Hui Yang is responsible for giving responses to all the feedback. 3. Replace the new LIPIDAG with the current version. List of Modifications to the LIPIDAG web site structure: 1. The current LIPIDAG site structure is not well organized, so a few changes have been made to set up a more reasonably organized one. 2. The new directory hierarchy is the following: newLIPIDAG-HUI ;the root directory for new LIPIDAG /newLIPIDAG List of Modifications to ASP Codes: (For Hui Yang ONLY) 1. Change the DATA SOURCE name to "newLIPIDAG", which opens the new LIPIDAG database. 2. Change the SQL Queries so they are consistent with the new LIPIDAG DB schema.
