A component of the ure gene cluster in E. coli, ureC, encodes a subunit of urease. We have investigated the distribution of ureC in 202 Shiga toxin-producing E. coli (STEC) strains from Austria belonging to 61 different serotypes. These strains were of human (n=150), animal (n=38), and food (n=14) origin. ureC was present in all 72 E. coli O157:H7 and O157:NM (non-motile) strains, as well as in all 29 strains of serotypes O26:H11/NM, O111:H8/NM and O145:NM. In contrast, none of eight sorbitol-fermenting E. coli O157:NM were ureC-positive. ureC occurred significantly more frequently among STEC that carry eae (113 of 132; 85.6%) than among eae-negative STEC strains (four of 70; 5.7%; p<0.0001). However, only 4 (2%) of the 202 strains (3.4% of ureC positive strains) expressed urease activity. There was no significant association (p=0.56) between urease expression and the source of the isolates (humans vs. animals). Nucleotide sequence analysis of PCR amplicons derived from all seven genes of the ure cluster in STEC of 10 different serotypes demonstrated a high degree of homology (>=99%), indicating a recent acquisition of not necessarily expressed ure genes. ... Blooms of the freshwater cyanobacterium Anabaena circinalis are recognised as an important health risk worldwide due to the production of a range of toxins such as saxitoxin (STX) and its derivatives, also known as paralytic shellfish poisoning (PSP) toxins. In this study the transcriptional profile of PSP toxin-producing and non-toxic strains of A. circinalis was investigated by means of a DNA microarray approach. Additionally, gene expression was studied after exposure of toxic A. circinalis cultures to lidocaine hydrochloride at 1 @mM for 2 h. Under standard growth conditions, a limited number of putative toxic-strain distinctive DNA fragments, identified in previous studies, were preferentially expressed in toxic versus non-toxic strains. The same genes did not significantly change their expression after exposure to 1 @mM lidocaine, conditions previously shown to induce STX production in the cyanobacterium Cylindrospermopsis raciborskii T3. Lidocaine supplementation, however, enhanced the transcription of genes involved in physiological adaptive responses and bloom formation in cyanobacteria, such as the gas vesicle structural protein A and phycocyanin. The heat shock protein HSP-70 and the chlorophyll-a binding protein isiA were significantly repressed by lidocaine exposure. Stress response proteins and genes implicated in secondary metabolism were repressed, including phosphopantetheinyl transferases. The BGGM^1 DNA microarray, used in this study, was shown to be suitable for gene expression studies in cultured toxic cyanobacteria and allowed the analysis of gene transcripts associated with surface scum formation by toxic A. circinalis. ... I would recomend this book to anyone looking for a comprehensive and easy to read text about toxic cyanobacteria.Chorus and Bartram have managed to fit together a great deal of very informative notes by many differentauthors in a format that progresses naturally from basic concepts ofcyanobacteria and their helpful/harmful effects to what can be done tomonitor, and manage them.Wheter you are just becoming interested incyanobacteria or lake health, or have been studying it for many years, thisbook should be part of your collection. The section on analytical methodsfor cyanobacteria provide excellent basic methodology in this area, butanyone wishing to undertake detailed analysis not from scratch would alsobe well off searching published methods in scientific journals as they willnot find all the nitty gritty details they will need.

A survey of residents in an area subject to annual toxic cyanobacterial blooms was undertaken to examine potential health effects of cyanobacteria toxins. The survey assessed the health of marine recreational water users in Deception Bay/Bribie Island area in northern Moreton Bay, Queensland, which is exposed to blooms of the nuisance and potentially harmful cyanobacterium Lyngbya majuscula. A postal survey was mailed to 5000 residents with a response rate of 27%. High numbers of people (78%) responding to the survey reported recreational water activity in Moreton Bay. Of those having marine recreational water activity, 34% reported at least one symptom after exposure to marine waters, with skin itching the most reported (23%). Younger participants had greater water exposure and symptoms than older participants. Participants with greater exposures were more likely to have skin and eye symptoms than less exposed groups, suggesting agents in the marine environment may have contributed to these symptoms. Of those entering Moreton Bay waters 29 (2.7%) reported severe skin symptoms, 12 of whom attended a health professional. Six (0.6%) reported the classic symptoms of recreational water exposure to L. majuscula, severe skin symptoms in the inguinal region. Participants with knowledge of L. majuscula were less likely to report less skin, gastrointestinal and fever and headache symptoms. In conclusion, high numbers of participants reported symptoms after exposure to waters subject to L. majuscula blooms but only a small number appeared to be serious in nature suggesting limited exposure to toxins. ...
